Principle of the Assay: The Human GITRL ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human GITRL in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Human GITRL coated on a 96-well plate. Standards and samples are pipetted into the wells and GITRL present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human GITRL antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of GITRL bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor(TNF) ligand family. It is mapped to 1q25.1. TNFSF18 is a ligand for receptor TNFRSF18/AITR/GITR. It has been shown to modulate T lymphocyte survival in peripheral tissues. This cytokine is also found to be expressed in endothelial cells, and is thought to be important for interaction between T lymphocytes and endothelial cells. TNFSF18-dependent modulation of tryptophan catabolism may represent an important mechanism of action of glucocorticoids, both physiologically and therapeutically.
