Background: Receptor tyrosine protein kinase (ErbB2) is also known as Neu and Her2, is a transmembrane glycoprotein in the ErbB family of tyrosine kinase and also belongs EGF receptors superfamily. ErbB2 is widely expressed in epithelial cells and over-expressed in lung carcinomas. Although HER2 itself lacks ligand binding domain and self inhibition to bind to growth factors, it can form heterodimer with EGF family having ligand binding domain function, which can stabilize ligand binding and enhance enzyme-mediated downstream molecular activation. The protease ADAM10 releases a 110 kDa soluble fragment of ErbB2 from the cell surface. ErbB2 plays important roles in cell growth, proliferation, differentiation and cancer. It was found that the ErbB2/ErbB3 heterodimer is expressed in the majority of tumor tissues. ErbB3, ErbB2 and neuregulin are all required for formation of the sympathetic nervous system.

Principle of the Assay: This ELISA kit is applied to the solid phase sandwich Enzyme Linked Immunosorbent Assay. The microtiter plate has been pre-coated with capture antibody. The antigen to be measured and the biotin-conjugated detection antibody are then added to the microtiter plate wells sequentially to form a capture antibody-antigen-detection antibody complex. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells, and then a TMB substrate solution is added to the wells to incubate. TMB substrate solution turns blue after the oxidation of HRP, and finally turns to yellow immediately after adding the stop solution. Color of TMB substrate positively correlated with total ErbB2 bound in the initial steps. The color is measured by spectrophotometrically with wavelength of 450nm. The concentration of ErbB2 in samples is then determined by comparing the O.D. of the samples to the standard curve.
