Specificity
This antibody strongly binds human and cynomolgus monkey PD-L1 and shows weak binding for mouse PD-L1. PD-L1 is highly expressed in many human tumors.. PD-L1 exerts an immune suppressive signal through binding to PD-1 and B7.1, and tumor expression of PD-L1 can mediate cancer immune evasion. Blocking this pathway, therefore provides a rationale for cancer immunotherapy.

The original antibody was generated by immunizing BALB/c mice with recombinant human PD-L1.
Preparation and Storage
Store at 4 degree C for up to 3 months. For longer storage, aliquot and store at -20 degree C.

FCM/FACS (Flow Cytometry)

(Flow cytometry using the Anti-PD-L1 antibody 332M7 (AAA72282). Paraformaldehyde fixed MDA-MB-231 cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody or the rabbit IgG version of 332M7 (AAA72282-23.0, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.)

Related Product Information for anti-PD-L1 antibody
The binding characterization of this antibody towards human, mouse and cynomolgus monkey PD-L1 was done using flow cytometry. This antibody was shown to block the binding of hPD-1-Fc protein to hPD-L1 expressed on the surface of the HEK-293T cells at a concentration of 10 ug/ml. The mouse version of this antibody was reported to inhibit patient derived melanoma tumor in tumor-bearing mice. There was also found an increase in the gene expression of CD8 and IFN-gamma in tumor samples from mice treated with mouse version of this antibody (US20180346571).
