Principle of the Assay: The LumiAb Human TNF-alpha Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human TNF-alpha concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human TNF-alpha while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human TNF-alpha or Tumor Necrosis Factor alpha, is a 233 amino acid cytokine protein encoded by the TNF gene located at locus 6p21.3 on chromosome 6. TNF-alpha binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR and is mainly secreted by macrophages, thus able to induce cell death of certain tumor cell lines. It is a potent pyrogen causing fever by direct action or by stimulation of IL-1 secretion and is implicated in the induction of cachexia. Under certain conditions, TNF-alpha is able to stimulate cell proliferation and induce cell differentiation. There are 2 main forms of TNF-alpha that are products of post-translational modifications. The soluble form derives from the membrane form by proteolytic processing. The membrane form, but not the soluble form, is phosphorylated on serine residues and dephosphorylation of the membrane form occurs by binding to soluble TNFRSF1A/TNFR1. Genetic variations in TNF are a cause of susceptibility psoriatic arthritis (PSORAS). PSORAS is an inflammatory, seronegative arthritis associated with psoriasis. It is a heterogeneous disorder ranging from a mild, non-destructive disease to a severe, progressive, erosive arthropathy. Five types of psoriatic arthritis have been defined: asymmetrical oligoarthritis characterized by primary involvement of the small joints of the fingers or toes; asymmetrical arthritis which involves the joints of the extremities; symmetrical polyarthritis characterized by a rheumatoid-like pattern that can involve hands, wrists, ankles, and feet; arthritis mutilans, which is a rare but deforming and destructive condition; arthritis of the sacroiliac joints and spine (psoriatic spondylitis).
