FCM/FACS (Flow Cytometry)

(Flow cytometric analysis of Human KIT(CD117) expression on TF-1 cells. Cells were stained with purified anti-Human KIT(CD117) (Filled hisgram), then a FITC-conjugated second step antibody. To demonstrate specificity of staining, the binding by 11996-R007 was blocked by preincubation of the purified antibody with 20ug recombinant human KIT(CD117) for 1 hour (Black solid line hisgram). The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.)

Related Product Information for anti-KIT antibody
C-Kit is a type 3 transmembrane receptor for MGF (mast cell growth factor, also known as stem cell factor). c-Kit contains 5 Ig-like C2-type (immunoglobulin-like) domains and 1 protein kinase domain. It belongs to the protein kinase superfamily, tyr protein kinase family, and CSF-1/PDGF receptor subfamily. C-Kit contains 5 Ig-like C2-type (immunoglobulin-like) domains and 1 protein kinase domain. C-Kit has tyrosine-protein kinase activity. Binding of the ligands leads to the autophosphorylation of KIT and its association with substrates such as phosphatidylinositol 3-kinase. Antibodies to c-Kit are widely used in immunohistochemistry to help distinguish particular types of tumor in histological tissue sections. It is used primarily in the identification of GISTs. In GISTs, c-Kit staining is typically cytoplasmic, with stronger accentuation along the cell membranes. C-Kit antibodies can also be used in the identification of mast cell tumors and in distinguishing seminomas from embryonal carcinomas. Mutations in the c-Kit gene are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism. Defects in KIT are a cause of acute myelogenous leukemia (AML). AML is a malignant disease in which hematopoietic precursors are arrested in an early stage of development. Note=Somatic mutations that lead to constitutive activation of KIT are detected in AML patients.

Product Notes

The KIT kit (Catalog #AAA255501) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Recombinant Anti-c-Kit Antibody, Rabbit Monoclonal reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's c-Kit can be used in a range of immunoassay formats including, but not limited to, IF (Immunofluorescence), ICC (Immunocytochemistry), FCM/FACS (Flow Cytometry). Researchers should empirically determine the suitability of the KIT kit for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "c-Kit, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
