(PRL (prolactin), also known as luteotropin, is a hormone secreted from the pituitary gland and is best known for its role in enabling mammals to produce milk. PRL plays an essential role in metabolism, regulation of the immune system through activating its specific membrane-anchored receptor (PRLR). A functional binding ELISA assay was conducted to detect the interaction of recombinant goat PRL and recombinant human PRLR. Briefly, PRL was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 ul were then transferred to PRLR-coated microtiter wells and incubated for 1h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PRL pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody for 1h at 37 degree C, wells were aspirated and washed 5 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50 uL stop solution to the wells and read at 450/630 nm immediately. The binding activity of recombinant goat PRL and recombinant human PRLR was shown in Figure 1, the EC50 for this effect is 0.1 ug/mL.)

Bioactivity
(CD320, also known as the Transcobalamin II receptor (TCb1R) and 8D6A, is an approximately 60 kDa transmembrane member of the LDL receptor family. CD320 is expressed in virtually all tissues and is tightly regulated according to the proliferative/differentiation status of the cell and the cellular levels of B12. Cellular uptake of vitamin B12 (cobalamin) requires capture of transcobalamin (TC) from the plasma by CD320. Cubilin (CUBN) is the intrinsic factor-vitamin B12 receptor, thus a functional binding ELISA assay was conducted to detect the interaction of recombinant human CD320 and recombinant human CUBN. Briefly, biotin-linked CD320 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100 ul were then transferred to CUBN-coated microtiter wells and incubated for 1h at 37 degree C. Wells were washed with PBST 3 times and incubation with Streptavidin-HRP for 30min, then wells were aspirated and washed 5 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50 ul stop solution to the wells and read at 450 nm immediately. The binding activity of recombinant human CD320 and recombinant human CUBN was shown in Figure 1, the EC50 for this effect is 0.4 ug/mL.)

(Western blot assay for Recombinant Mononucleosomes H3.1 (K4I) 2 ug Recombinant Mononucleosomes H3.1 (K4I) were incubated with 0 or 10 nM MLL2 Complex respectively in reaction buffer containing 50 mM Tris-HCl pH 8.6, 0.02% Triton X-100, 2 mM MgCl2, 1 mM TCEP and 50 uM SAM for 3 hr at room temperature. Half of the reactions were run on a 12.5% SDS-PAGE gel and detected with H3K4me2 antibody and anti-H4 antibody , respectively. H4 was detected as loading control. Recombinant Mononucleosomes H3.1 and Histone H3.1 was used as a positive control. The result shows that no H3K4me2 products were generated when Mononucleosomes H3.1 (K4I) were incubated with MLL2 Complex.)
