(Glypican 1 (GPC1) is one of the six glycosylphosphatidylinositol-anchored, cell surface heparan sulfate proteoglycans that acts as a growth factor signaling. Cells known to express GPC-1 include neurons, smooth and skeletal muscle cells, keratinocytes, osteoblasts, Schwann cells, immature dendritic cells, and tumor, plus tumor-associated vascular endothelial cells. The function of GPC-1 is complex and varied, it can modulate various signaling pathways including Heparin Binding Epidermal Growth Factor Like Growth Factor (HBEGF), fibroblast growth factors (FGF), vascular endothelial growth factor-A (VEGF-A), transforming growth factor-beta (TGF-beta), Wnt, Hedgehog (Hh), and bone morphogenic protein (BMP) through specific interactions with pathway ligands and receptors. Besides, Syndecan 4 (SDC4) has been also identified as an interactor of GPC1), thus a functional binding ELISA assay was conducted to detect the interaction of recombinant mouse GPC1 and recombinant chicken SDC4. Briefly, GPC1 was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 ul were then transferred to SDC4-coated microtiter wells and incubated for 1h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-GPC1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody for 1h at 37 degree C, wells were aspirated and washed 5 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50 uL stop solution to the wells and read at 450/630 nm immediately. The binding activity of recombinant mouse GPC1 and recombinant chicken SDC4 was shown in Figure 1, the EC50 for this effect is 0.04 ug/mL.)

(Galectin-12 is a member of a family of mammalian lectins known as galectins. The galectins constitute a large family of carbohydrate-binding proteins that function in many systems both intracellularly and following secretion. Galectins contain either one or two carbohydrate recognition domains (CRR) which mediate recognition of N-acetyl-lactosamine-containing glycoproteins. Individual galectins differ in their tissue distribution and in their carbohydrate-binding specificities. Galectin-12 is predominantly expressed in adipose tissue and detected also in macrophages and other leukocytes. It plays an important role in cell-cell adhesion, cell-matrix interactions, macrophage activation, angiogenesis, metastasis, apoptosis. In this case, we chose rabbit erythrocyte (RaE) to assay its ability of agglutination. A general procedure for hemagglutination assay (or haemagglutination assay; HA) is as follows, two-fold dilute the recombinant Po GAL12 with 0.9% sodium chloride injection, add 50uL a serial dilution of GAL12 to each well of a U or V-bottom shaped 96-well microtiter plate. The final well serves as a negative control with no GAL12, replace with 50uL 0.9% sodium chloride injection. Then add 50uL 1% rabbit erythrocyte to each well and mixed gently. The plate is incubated for 3 hours at room temperature. The results are shown in Figure 1. It was obvious that the minimal effective concentration of GAL12 is 0.195 ug/mL.)

(VCP is an ATPase Associated with diverse cellular Activities (AAA) that has a key role in maintaining cellular homeostasis. VCP is ubiquitously expressed in tissues. At cellular level, VCP functions in many cellular compartments; VCP localizes mainly in the cytoplasm, while a smaller fraction binds to organelles or localizes in the nucleus, where it is implicated in different pathways. VCP assembles and acts as a homo-hexamer, in which each monomer is structured in an N-terminal domain that interacts with adaptors and co-factors; in two ATPase domains, D1 and D2, that by hydrolyzing ATP concur, respectively, in the hexamer formation and in supporting VCP activity; and in a C-terminal domain that binds to a small subset of co-factors and adaptors cooperating with D2 activity. The main role of VCP is to recognize and to extract ubiquitinated proteins from membranes, protein complexes, protein aggregates, or chromatin, modulating their ubiquitination or de-ubiquitination, and to enhance their degradation through the UPS or the autophagic pathway. The activity of recombinant human VCP was measured by its ability to hydrolyze the substrate ATP to phosphate which was detected by the malachite green phosphate detection Kit (Beyotime # S0196M). The reaction was performed in the assay buffer 20 mM HEPES, 13 mM NaCl and 1% glycerol, pH 8.5, initiated by addition 27 uL of various concentrations of VCP (diluted by assay buffer) to 3 uL of 100 mM ATP and 100 mM MgCl2 mixture. The final well serves as a negative control with no VCP, replaced with 27 uL assay buffer. Incubated at 37 degree C for 5min, then stop the reaction by quickly freezing samples in the dry ice and ethanol. 4 ul samples was added to 196 ul assay buffer after thawing at room temperature followed by adding 70 ul phosphate detection agent and incubated at room temperature for 30min, then read at a wavelength of 630 nm. (Complement Component 3 (C3) is a vital component of the complement system that plays a central role in immune response and inflammation. C3 is a beta? globulin synthesized by the liver, whose structure consists of two polypeptide chains, alpha and beta, and is the most abundant complement component in serum. It's been identified that C3 can interact with Complement Factor B (CFB), thus a functional binding ELISA assay was conducted to detect the interaction of recombinant human C3 and recombinant mouse CFB. Briefly, CFB was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 ul were then transferred to C3-coated microtiter wells and incubated for 1h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-CFB pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody for 1h at 37 degree C, wells were aspirated and washed 5 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50 uL stop solution to the wells and read at 450/630 nm immediately. When Recombinant C3 is immobilized at 2 ug/mL (100 uL/well), the concentration of CFB that produces 50% optimal binding response is found to be approximately 5.96 ug/mL.)

(Carbonic Anhydrase (CA) catalyzes the reversible reaction of CO2 H2O = HCO3- H, which is fundamental to many processes such as respiration, renal tubular acidification and bone resorption. CA13 is a cytosolic enzyme with a unique and widespread distribution pattern as compared to the other cytosolic CAs, indicating that it may play important physiological roles in several organs. The activity of recombinant human CA13 was measured by its ability to hydrolyze 4-Nitrophenyl acetate (4-NPA) to 4-Nitrophenol. The reaction was performed in 12.5 mM Tris, 75 mM NaCl, pH 7.5 (assay buffer), initiated by addition 50 uL of various concentrations of CA13 (diluted by assay buffer) to 50 uL of 2 mM substrate 4-NPA (100 mM stock in Acetone, diluted by assay buffer).
