(Western blot assay for Recombinant Mononucleosomes H3.1 (K9M) 0.5 ug Recombinant Histone H3.1 (K9M) which was used for assembly of nucleosomes was incubated with 0 or 5 nM G9A respectively in reaction buffer containing 50 mM Tris-HCl pH 8.6, 0.02% Triton X-100, 2 mM MgCl2, 1 mM TCEP and 50 uM SAM for 3 hr at room temperature. Half of the reactions were run on a 12.5% SDS-PAGE gel and detected with H3K9me2 antibody . Histone H3.1 was used as a control. The Western Blot result shows that only the K9 of histone H3.1 can be methylated by G9A.)

Western blot analysis of AMPK alpha 1 using anti-AMPK alpha 1 antibody (AAA124608).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: human PANC-1 whole cell lysates,Lane 2: human Jurkat whole cell lysates.After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-AMPK alpha 1 antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for AMPK alpha 1 at approximately 63KD. The expected band size for AMPK alpha 1 is at 63KD.)
