product-image-AAA150082_BIOACTIVITY10.jpg Bioactivity (Ribonuclease P (RNASEP) is a type of ribonuclease which cleaves RNA. RNase P is unique from other RNases in that it is a ribozyme – a ribonucleic acid that acts as a catalyst in the same way that a protein based enzyme would. Its function is to cleave off an extra, or precursor, sequence of RNA on tRNA molecules. Besides, Methyl CpG Binding Protein 2 (MECP2) has been identified as an interactor of RNASEP, thus a binding ELISA assay was conducted to detect the interaction of recombinant human RNASEP and recombinant human MECP2. Briefly, RNASEP were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100L were then transferred to MECP2-coated microtiter wells and incubated for 2h at 37. Wells were washed with PBST and incubated for 1h with anti-RNASEP pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of RNASEP and MECP2 was shown in Figure 1, and this effect was in a dose dependent manner.Figure. product-image-AAA150135_BIOACTIVITY10.jpg Bioactivity (Figure. The binding activity of GDNF with GFRa2.Glial cell-derived neurotrophic factor (GDNF) is a small protein that potently promotes the survival of many types of neurons. This protein was shown to promote the survival and differentiation of dopaminergic neurons in culture, and was able to prevent apoptosis of motor neurons induced by axotomy. GDNF is processed to a mature secreted form that exists as a homodimer. The mature form of the protein is a ligand for the product of the RET (rearranged during transfection) protooncogene. Besides, Glial Cell Line Derived Neurotrophic Factor Receptor Alpha 2 (GFRa2) has been identified as an interactor of GDNF, thus a binding ELISA assay was conducted to detect the interaction of recombinant human GDNF and recombinant human GFRa2. product-image-AAA161908_BIOACTIVITY13.jpg Bioactivity (Lysophosphatidic acid phosphatase type 6 (ACP6), is a 44-47 kDa monomeric member of the histidine acid phosphatase family of proteins. ACP6 is highly expressed in kidney, heart, small intestine, muscle, liver, prostate, testis, ovary and exists as two isoforms as a result of alternative splicing events. It plays a role in regulating lipid metabolism that hydrolyzes LPA to monoacylglycerol (MAG) and phosphate. Annexin V (ANXA5) binds to phospholipid bilayers, forming two-dimensional crystals that block the phospholipids from availability for coagulation enzyme reactions. A functional binding ELISA assay was conducted to detect the interaction of recombinant human ACP6 and recombinant mouse ANXA5. Briefly, ACP6 was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 ul were then transferred to ANXA5-coated microtiter wells and incubated for 1h at 37 degree C. product-image-AAA161671_BIOACTIVITY13.jpg Bioactivity (Insulin-like growth factor 2 (IGF2) is one of three protein hormones that share structural similarity to insulin. It has growth-regulating, insulin-like and mitogenic activities. IGF2 exerts its effects by binding to the IGF-1 receptor and to the short isoform of the insulin receptor. IGF2 may also bind to the IGF2 receptor (also called the cation-independent mannose 6-phosphate receptor), which acts as a signalling antagonist; that is, to prevent IGF2 responses. Besides, Insulin Like Growth Factor Binding Protein 2 (IGFBP2) has been identified as an interactor of IGF2, thus a functional binding ELISA assay was conducted to detect the interaction of recombinant mouse IGF2 and recombinant mouse IGFBP2. Briefly, IGF2 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100 ul were then transferred to IGFBP2-coated microtiter wells and incubated for 2h at 37 degree C. product-image-AAA161690_BIOACTIVITY13.jpg Bioactivity (Oncostatin M (OSM) is an approximately 30 KDa secreted cytokine belonging to the Interleukin-6 family. Like other members of the IL-6 family such as IL-11, CNTF, and Cardiotrophin-1, OSM plays crucial roles in inflammation, neuroprotection, hematopoiesis, metabolism and development. To test the effect of OSM on cell proliferation, TF-1 cells were seeded into triplicate wells of 96-well plates at a density of 20,000 cells/well with various concentrations of recombinant human OSM. After incubated for 72 hours, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8(CCK-8). Briefly, 10 ul of CCK-8 solution was added to each well of the plate, then the absorbance at 450 nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of TF-1 cells after incubation with OSM for 72 hours observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8) assay after incubation with recombinant human OSM for 72 hours. The result was shown in Figure 2. It was obvious that recombinant human OSM significantly increased cell viability of TF-1 cells, the EC50 was 0.08 ng/ml.)

product-image-AAA161695_BIOACTIVITY13.jpg Bioactivity (Tissue inhibitors of metalloproteinases or TIMPs are a family of proteins that regulate the activation and proteolytic activity of the zinc enzymes known as matrix metalloproteinases (MMPs). There are four members of the family, TIMP-1, TIMP-2, TIMP-3, and TIMP-4. TIMP-2 is a non N-glycosylated protein with a molecular mass of 22 kDa produced by a wide range of cell types, which inhibits MMPs non-covalently by the formation of binary complexes. TIMP-2 also has erythroidpotentiating and cell growth promoting activities. The activity of recombinant bovine TIMP2 was measured by its ability to inhibit rhMMP2 cleavage of a fluorogenic peptide substrate MCA-Pro-Leu-Gly-Leu-DPA-Ala-Arg-NH2 in the assay buffer 50 mM Tris, 10 mM CaCl2, 150 mM NaCl, 0.05% (w/v) Brij-35, pH 7.5. rhMMP2 was diluted to 100 ug/ml and activated with 1 mM APMA at 37 degree C for 1 hour and rbTIMP2 (MW: 51.75 KD) was diluted to different concentrations with the assay buffer. Mix 8 ul of rhTIMP2 curve dilutions, 12.8 ul of activated rbMMP-2, and 59.2 ul of assay buffer, including a control containing assay buffer and the diluted rhMMP-2 and incubate the reactions for 2 hours at 37 degree C. Loading 50 ul of the incubated mixtures which were diluted five-fold in assay buffer into empty wells of a plate, and start the reaction by adding 50 ul of 20 uM substrate. Include a substrate blank containing 50 ul of assay buffer and 50 ul of 20 uM substrate. Then read at excitiation and emission wavelengths of 320 nm and 405 nm, respectively, in kinetic mode for 5 minutes. The result was shown in Figure 1 and it was obvious that recombinant bovine TIMP2 significantly decreased rhMMP2 activity. The inhibition IC50 was )

product-image-AAA161738_BIOACTIVITY13.jpg Bioactivity (Apolipoprotein A1 (APOA1) is the major protein component of HDL particles in plasma. It is a cofactor for lecithin cholesterolacyltransferase (LCAT) which is responsible for the formation of most plasma cholesteryl esters. ApoA1 was also isolated as a prostacyclin (PGI2) stabilizing factor, and thus may have an anticlotting effect. ApoA1 is often used as a biomarker for prediction of cardiovascular diseases. Besides, Plasminogen Activator, Urokinase (uPA) has been identified as an interactor of APOA1, thus a functional binding ELISA assay was conducted to detect the interaction of recombinant human APOA1 and recombinant human uPA. product-image-AAA161867_BIOACTIVITY13.jpg Bioactivity (Cathepsin S (CTSS) is a lysosomal cysteine protease of the papain family. It plays a major role in the processing of the MHC class IIassociated invariant chain. It has been implicated in the pathogenesis of several diseases such as Alzheimer's disease and degenerative disorders associated with the cells of the mononuclear phagocytic system. Mouse Cathepsin S is synthesized as a preproenzyme of 340 amino acid residues consisting a signal peptide (residues 117), a pro region (residues 18122), and the mature enzyme (residues 123340). Cathepsin S is less abundant in tissues than Cathepsins B, L and H. The activity of recombinant mouse CTSS is measured by its ability to cleave a fluorogenic peptide substrate Mca-Arg-Pro-Lys-Pro-Val-Glu-Nval-Trp-Arg-Lys(Dnp)-NH2 in the assay buffer 50 mM NaOAc, 5 mM DTT, 250 mM NaCl, pH 4.5. The rmCTSS is diluted to 50 ug/ml in assay buffer, then incubated at room temperature for 2h. The incubated rmCTSS is diluted to 5 ug/mL in assay buffer. Loading into a black well plate 50 uL of 5 ug/mL rmCTSS and start the reaction by adding 50 uL of 20 uM substrate, with a substrate blank containing 50 uL assay buffer, 50 uL substrate, and no rmCTSS. Then read at excitiation and emission wavelengths of 320 nm and 405 nm, respectively, in kinetic mode for 5 minutes. The specific activity of recombinant mouse CTSS is > 10 pmol/min/ug.)

product-image-AAA161874_BIOACTIVITY13.jpg Bioactivity (Microsomal human cytochrome P450 17A1 (CYP17A1, 17alpha-hydroxylase, 17,20-lyase) belongs to the cytochrome P450 super family, is expressed in the adrenals and gonads, with minor amounts in the brain, placenta, and heart. CYP17A1 enzyme operates at a key juncture of human steroidogenesis, controlling the levels of mineralocorticoids influencing blood pressure, glucocorticoids involved in immune and stress responses, and androgens and estrogens involved in development and homeostasis of reproductive tissues. Receptor Tyrosine Protein Kinase erbB-2 (ErbB2) has been identified as an interactor of CYP17A1, thus a functional binding ELISA assay was conducted to detect the interaction of recombinant human CYP17A1 and recombinant human ErbB2. Briefly, CYP17A1 was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 ul were then transferred to ErbB2-coated microtiter wells and incubated for 1h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-CYP17A1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody for 1h at 37 degree C, wells were aspirated and washed 5 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50 uL stop solution to the wells and read at 450/630 nm immediately. The binding activity of recombinant human CYP17A1 and recombinant human ErbB2 was shown in Figure 1, the EC50 for this effect is 0.03 ug/mL.)

product-image-AAA161807_BIOACTIVITY13.jpg Bioactivity (Retinol-binding protein 4 (RBP4) is the specific carrier for retinol (also known as vitamin A), and is responsible for the conversion of unstable and insoluble retinol in aqueous solution into stable and soluble complex in plasma through their tight interaction. As a member of the lipocalin superfamily, RBP4 containing a beta-barrel structure with a well-defined cavity is secreted from the liver, and in turn delivers retinol from the liver stores to the peripheral tissues. In plasma, the RBP4-retinol complex interacts with transthyretin (TTR), and this binding is crucial for preventing RBP4 excretion through the kidney glomeruli. RBP4 expressed from an ectopic source efficiently delivers retinol to the eyes, and its deficiency affects night vision largely. Recently, RBP4 as an adipokine, is found to be expressed in adipose tissue and correlated with obesity, insulin resistance (IR) and type 2 diabetes (T2DM). The activity of recombinant pig RBP4 was measured by its ability to bind all-trans retinoic acid. The binding of retinoic acid results in the quenching of Trp fluorescence in RBP4. RBP4 was diluted to 50 ug/ml in 50 mM Tris, 10 mM CaCl2, 150 mM NaCl, pH 7.5 (assay buffer) and the retinoic acid was diluted to 400, 200, 100, 30, 10, 3, 1 and 0.5 uM in 95% ethanol. Mixing 112.5 uL of 50 ug/mL rpRBP4 and 12.5 uL of retinoic acid serial dilutions in microtubes and a blank containing 112.5 uL of 50 ug/mL rpRBP4 and 12.5 uL of 95% ethanol, then incubate at room temperature for 30 minutes. Loading 100 ul of the reaction mixtures and blank and read at excitation and emission wavelengths of 280 nm and 340 nm (top read), respectively, in endpoint mode. The result was shown in figure 1, the 50% binding concentration (BC50) is >30 uM.)

Thioflavin T is a fluorescent dye that binds to beta sheet-rich structures, such as those in alpha synuclein fibrils. Upon binding, the emission spectrum of the dye experiences a red-shift and increased fluorescence intensity. Thioflavin T emission curves show increased fluorescence (correlated to alpha synuclein protein aggregation) over time when 10 uM of Type 1 alpha synuclein preformed fibrils is combined with 100 uM of alpha synuclein monomer , as compared to when 10 uM of Type 2 alpha synuclein preformed fibrils is combined with 100 uM of alpha synuclein monomer or 100 uM of alpha Synuclein monomer . What Are Active Proteins?

Proteins are large molecules made up of long chains of amino acids.
They will typically fold into a very particular 3-dimensional shape/conformation, that is sometimes referred to as their “native” form, which allows them to work properly in the body. For the purposes of product categorization, AAA Biotech will typically refer to proteins purified from their original animal host as being “native” proteins (this is to signify their difference compared to their “recombinant” or “synthetic” protein counterparts).

If a protein successfully folds into the correct shape, it is will typically display high fidelity characteristics to its original protein in its original animal host, and be classified as an active protein, as it will be able to function “normally” in most enzymatic or binding capacities. If it loses this shape, due to factors such as heat or strong chemicals (such as detergents), it becomes inactive and is no longer able to perform its basic functions. All of the proteins in this category are made under strict quality control, and they are active, pure, low in contaminants, and stable.
Most are stored as freeze-dried powders and come without extra tags, so they’re very close to the actual natural/native form.
