(Protein Z-dependent Protease Inhibitor (ZPI), also known as SerpinA10 (SERine Proteinase INhibitor-clade A10) is a monomeric, secreted member of the A (or extracellular) clade within the serpin superfamily of protease inhibitors. In general, members of this superfamily regulate multiple proteolytic cascades, and are particularly effective due to the fact that their inhibitory activities can be fine-tuned through the participation of discrete, non-serpin co-factors. Serpins are unusual in that they are one-time use, non-recyclable proteins whose native state is thermodynamically unstable. The activity of recombinant human SERPINA10 was measured by its ability to inhibit Coagulation Factor X cleavage of a fluorogenic peptide substrate Mca-RPKPVE-Nval-WRK(Dnp)-NH2 in the assay buffer 50 mM Tris, 10 mM CaCl2, 150 mM NaCl, 0.05% (w/v) Brij-35, pH 7.5. Coagulation Factor X was diluted to 5 ug/ml in the assay buffer and 25 ul different concentrations of recombinant human SERPINA10 (MW: 78.26 KD) was incubated with 25 ul diluted Coagulation Factor X at 37 degree C for 30 minutes. Loading 50 uL 20 uM substrate to start the reaction including a substrate blank containing 50 uL of assay buffer and 50 uL of 20 uM substrate. Then read at excitiation and emission wavelengths of 320 nm and 405 nm, respectively, in kinetic mode for 5 minutes. (Parathyroid hormone (PTH) is an 84-amino-acid peptide hormone that is secreted by the parathyroid gland. PTH plays an osteogenic role by acting directly on mesenchymal stem cells, cells with an osteoblastic lineage, osteocytes, and T cells. It also participates as an osteoclast by indirectly acting on osteoclast precursor cells and osteoclasts and directly acting on T cells. PTH is major endocrine regulators of vitamin D metabolism. It proves, with data, this hormone a potent down-regulator of VDR. A functional binding ELISA assay was conducted to detect the interaction of recombinant human PTH and recombinant mouse VDR. Briefly, PTH was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 ul were then transferred to VDR-coated microtiter wells and incubated for 1h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PTH pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody for 1h at 37 degree C, wells were aspirated and washed 5 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50 uL stop solution to the wells and read at 450/630 nm immediately. (Granulocyte-macrophage colony-stimulating factor (GM-CSF), also known as colony stimulating factor 2 (CSF2), is a monomeric glycoprotein secreted by macrophages, T cells, mast cells, natural killer cells, endothelial cells and fibroblasts that functions as a cytokine. GM-CSF is a monomeric glycoprotein that functions as a cytokine - it is a white blood cell growth factor. It also has some effects on mature cells of the immune system, such as inhibiting neutrophil migration and causing an alteration of the receptors expressed on the cells surface. GM-CSF signals via signal transducer and activator of transcription, STAT5. In macrophages, it has also been shown to signal via STAT3. Besides, Colony Stimulating Factor 2 Receptor Alpha (CSF2Ra) has been identified as an interactor of GM-CSF, thus a binding ELISA assay was conducted to detect the interaction of recombinant human GM-CSF and recombinant human CSF2Ra. Briefly, GM-CSF were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100 ul were then transferred to CSF2Ra-coated microtiter wells and incubated for 1h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-GM-CSF pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50 uL stop solution to the wells and read at 450 nm immediately. The binding activity of recombinant human GM-CSF and recombinant human CSF2Ra was shown in Figure 1, the EC50 for this effect is 0.04 ug/mL.)
