product-image-AAA150057_AD11.jpg Application Data (Interleukin 11 (IL-11) is a multifunctional cytokine. It is a key regulator of multiple events in hematopoiesis, most notably the stimulation of megakaryocyte maturation. IL-11 has been demonstrated to improve platelet recovery after chemotherapy-induced thrombocytopenia, induce acute phase proteins, modulate antigen-antibody responses, participate in the regulation of bone cell proliferation and differentiation IL-11 causes bone-resorption. Besides, IL-11 have been proved can promote migration of A549 cells, 5×104 cells were seeded into 6 well plates. After cell confluent, replace with DMEM without serum overnight. Using a (yellow) pipette tip make a straight scratch, simulating a wound, then washing the wells three times with PBS. Adding 1% serum standard DMEM containing various concentrations of recombinan human IL-11 to each well, incubating the plate for 48 hours at 37, 5% CO2. The results observed by inverted microscope was shown in Figure.(A) A549 cells cultured in DMEM with 100ng/mL IL-11 for 48h; (B) A549 cells cultured in DMEM before addition IL-11.Figure. product-image-AAA150092_AD10.png Application Data (Interleukin 1 beta (IL-1) also known as leukocytic pyrogen, leukocytic endogenous mediator, mononuclear cell factor, lymphocyte activating factor and other names, is a member of the interleukin 1 family of cytokines. This cytokine is an important mediator of the inflammatory response, and is involved in a variety of cellular activities, including cell proliferation, differentiation, and apoptosis. It has been reported that IL-1 can induced IL-8 production in A549 cells. To test the bioactivity of IL-1, A549 cells were seeded into 24-well plate at a density of 1x106 cells/mL, and allowed to attach overnight before treated with certain concentrations (0.1ng/mL-1000ng/mL) of IL-1 for 24h and IL-8 levels in the cell supernatant were determined by ELISA. IL-8 levels in the cell supernatant of A549 cells increased significantly after stimulated with IL-1 have shown in Figure1.Figure. IL-8 levels in the cell supernatant of A549 induced by IL-1.)

product-image-AAA150108_AD11.jpg Application Data (Figure. Cell proliferation of Raji cells after stimulated with GP39.Glycoprotein 39, Cartilage (GP39) also known as Chitinase-3-like protein 1 (CHI3L1) is a secreted glycoprotein that is approximately 40kDa in size that in humans is encoded by the CHI3L1 gene. GP39 plays a role in cancer cell proliferation, survival, invasiveness and in the regulation of cell-matrix interactions. To test the effect of GP39 on cell proliferation, Raji cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well with 1% serum standard 1640 which contains various concentrations of recombinant human GP39. After incubated for 5 days, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37. Proliferation of Raji cells after incubation with GP39 for 5 days observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8) assay after incubation with recombinant GP39 for 5 days. The result was shown in Figure 2. It was obvious that GP39 significantly increased cell viability of Raji cells.(A) Raji cells cultured in 1640, stimulated with 10ng/mL GP39 for 5 days; (B) Unstimulated Raji cells cultured in 1640 for 5 days.)

product-image-AAA152173_BIOACTIVITY13.png Bioactivity (The galectins constitute a large family of carbohydrate-binding proteins with specificity for N-acetyl-lactosamine-containing glycoproteins. At least 14 mammalian galectins, which share structural similarities in their carbohydrate recognition domains (CRD), have been identified. The galectins have been classified into the prototype galectins (-1, -2, -5, -7, -10, -11, -13, -14), which contain one CRD and exist either as a monomer or a noncovalent homodimer; the chimera galectins (Galectin-3) containing one CRD linked to a nonlectin domain; and the tandem-repeat galectins (-4, -6, -8, -9, -12) consisting of two CRDs joined by a linker peptide. Galectin-7 may also be involved in cell-cell and cell-matrix interactions and exogenous galectin has been found to accelerate the re-epithelialization of woundsIt can agglutinate red blood. In this case, we chose rabbit erythrocyte (RaE) to assay its ability of agglutination. A general procedure for hemagglutination assay (or haemagglutination assay; HA) is as follows, two-fold dilute the recombinant Ra GAL7 with 0.9% sodium chloride injection, add 50uL a serial dilution of GAL7 to each well of a U or V-bottom shaped 96-well microtiter plate. The final well serves as a negative control with no GAL7, replace with 50uL 0.9% sodium chloride injection. Then add 50uL 1% rabbit erythrocyte to each well and mixed gently. The plate is incubated for 3 hours at room temperature. The results are shown in Figure 1. It was obvious that the minimal effective concentration of GAL7 is 25 ug/mL.)

product-image-AAA161730_BIOACTIVITY13.jpg Bioactivity (Human TFPI, also known as lipoprotein-associated coagulation inhibitor (LACI) and extrinsic pathway inhibitor (EPI), is a physiological inhibitor of extrinsic pathway of coagulation and has biological functions of anticoagulation and anti-inflammation. It is a secreted protein with a N-terminal acidic region, three Kunitz (K) domains separated with by two linker regions, and a C-terminal basic region. The activity of recombinant human TFPI was measured by its ability to inhibit trypsin cleavage of a fluorogenic peptide substrate Mca-RPKPVE-Nval-WRK(Dnp)-NH2 in the assay buffer 50 mM Tris, 10 mM CaCl2, 150 mM NaCl, 0.05% (w/v) Brij-35, pH 7.5. Trypsin was diluted to 50 ug/ml in the assay buffer and 20 ul different concentrations of recombinant human TFPI (MW: 61.9 KD) was incubated with 20 ul diluted trypsin at 37 degree C for 15 minutes. Loading 50 uL of the incubated mixtures which were diluted five-fold in assay buffer into empty wells of a plate, and start the reaction by adding 50 uL of 20 uM substrate. Include a substrate blank containing 50 uL of assay buffer and 50 uL of 20 uM substrate. Then read at excitiation and emission wavelengths of 320 nm and 405 nm, respectively, in kinetic mode for 5 minutes. What Are Active Proteins?

Proteins are large molecules made up of long chains of amino acids.
They will typically fold into a very particular 3-dimensional shape/conformation, that is sometimes referred to as their “native” form, which allows them to work properly in the body. For the purposes of product categorization, AAA Biotech will typically refer to proteins purified from their original animal host as being “native” proteins (this is to signify their difference compared to their “recombinant” or “synthetic” protein counterparts).

If a protein successfully folds into the correct shape, it is will typically display high fidelity characteristics to its original protein in its original animal host, and be classified as an active protein, as it will be able to function “normally” in most enzymatic or binding capacities. If it loses this shape, due to factors such as heat or strong chemicals (such as detergents), it becomes inactive and is no longer able to perform its basic functions. All of the proteins in this category are made under strict quality control, and they are active, pure, low in contaminants, and stable.
Most are stored as freeze-dried powders and come without extra tags, so they’re very close to the actual natural/native form.

FAQ

1. What are active proteins used for in research?

Active proteins are used primarily in the study of how proteins function, in characterizing/discovering drug interactions, supporting cell growth, running biochemical assays, and in development of diagnostics or therapeutics.

2. How are AAA Biotech's active proteins validated?

AAA Biotech’s active proteins are validated through strict quality control and functional assays to ensure they are properly folded and active. “Active”, though, can be an ambiguous term, so if a specific “activity” or “binding” capability of a protein is of crucial interest to you, please inquire with us prior to purchase, and we will provide further details on how the “Active” modifier was determined to be applicable.

3. Are these proteins tested for biological activity?

Yes, all active proteins from AAA Biotech are tested to confirm they have the expected biological activity before being offered for use. Though, said “biological activity” can be either “enzymatic”, “binding”, or both.
