product-image-AAA161827_BIOACTIVITY13.jpg Bioactivity (SP-D (surfactant protein-D) is a 43 kDa member of the collectin family of innate immune modulators. It is constitutively secreted by alveolar lining cells and epithelium associated with tubular structures. Its principal components consist of a collagen-like region and a C-terminal carbohydrate recognition domain (CRD), a structure that further places it in a subset of an expanded group of proteins termed defense collagens. SP-D is known to bind both SIRP alpha and the calreticulin/CD91 complex on macrophages. When the ratio of antigen/pathogen to available CRDs is low, antigen can be bound without occupying all available CRDs. The free CRDs will bind to SIRP alpha, generating a signal that downmodulates the inflammatory response. When virtually all CRDs are occupied by ligand, however, free CRDs are not available for SIRP alpha binding. The activity of the recombinant human SPD was measured by its ability to bind fluorescein-conjugated E. coli bioparticles. The rhSPD was diluted to 10 ug/ml in assay buffer of 20 mM Tris, 137 mM NaCl, 1 mM CaC12, pH7.4 and fluorescein-conjugated E. coli was diluted to 7*108 cells/ml. Equal volume of 10 ug/ml rhSPD and fluorescein-conjugated E. coli were mixed and incubated at room temperature for 1h. 10 ul mixture was took onto the slide and observed under the fluorescence microscopy. The result was shown in figure 1, the rhSPD could bind fluorescein-conjugated E. coli bioparticles.)

product-image-AAA161830_BIOACTIVITY13.jpg Bioactivity (Four distinct genes encode alkaline phosphatases (APs) in humans. The ALPL gene encodes the liver/bone/kidney isozyme, also known as the tissuenonspecific AP (TNAP). In comparison, ALPI, ALPP and ALPPL2 encode intestinal, placental and placental-like or germ cell APs, respectively. The serum levels of human APs are useful tumor markers. There are many mutations in the ALPL gene, leading to different forms of hypophosphatasia, characterized by poorly mineralized cartilage and bones. The native ALPL is a glycosylated homodimer attached to the membrane through a GPI-anchor. The activity assay of recombinant mouse ALPL was measured by its ability to cleave a peptide substrate, 4-Methumbelliferly phosphate. The reaction was performed in 50 mM Tris, 1 mM MgCl2, pH 9.0 (assay buffer), ainitiated by addition 50 ul of 0.3 ug/ml ALPL (diluted by assay buffer) to 50 uL of 50 uM substrate. Read at excitation and emission wavelengths of 365 nm and 445 nm (top read), respectively, in kinetic mode for 5 minutes. The specific activity of recombinant mouse ALPL is >11000 pmol/min/ug.)

product-image-AAA161875_BIOACTIVITY13.jpg Bioactivity (Growth Differentiation Factor 15 (GDF-15), also called Macrophage Inhibitory Cytokine 1 (MIC-1), is a divergent member of the Transforming Growth Factor beta (TGF-beta) superfamily. Human GDF-15 shares 66% and 68% amino acid sequence identity with the rat and mouse proteins, respectively. GDF-15 is highly expressed in placenta and brain, and it is expressed at lower levels in kidney, pancreas, prostate, and colon. Similar to other TGF-beta family proteins, GDF-15 is synthesized as a large precursor protein that is cleaved at a dibasic cleavage site (RxxR) to release the mature protein. Biologically active GDF-15 is a disulfide-linked homodimer of the mature protein. GDF15 has been shown to have various functions, including inhibition of Tumor Necrosis Factor alpha (TNF-alpha) production from lipopolysaccharide-stimulated macrophages and the induction of cartilage formation. A functional binding ELISA assay was conducted to detect the interaction of recombinant rat GDF-15 and recombinant human GP9. Briefly, GDF-15 was diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100 ul were then transferred to GP9-coated microtiter wells and incubated for 1h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-GDF-15 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody for 1h at 37 degree C, wells were aspirated and washed 5 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50 uL stop solution to the wells and read at 450/630 nm immediately. The binding activity of GDF-15 and GP9 was shown in Figure 1, the EC50 for this effect is 0.47 ug/mL.)

product-image-AAA161882_BIOACTIVITY13.jpg Bioactivity (Amine Oxidase, Copper Containing 3 (AOC3) is a copper amine oxidase with a topaquinone cofactor. AOC3 is a Type II integral membrane protein, but a soluble form of the enzyme is present in human serum, and its level increases in diabetes and some inflammatory liver diseases. AOC3 catalyzes the oxidative deamination of small primary amines such as methylamine, benzylamine, and aminoacetone in a reaction that produces an aldehyde, ammonia, and H2O2. The enzyme is sensitive to inhibition by semicarbazide. AOC3 expression is highest in the endothelium of lung, heart, and intestine, but low in tissues such as brain, spleen, kidney, and liver. The activity of recombinant human AOC3 was measured by its ability to produce hydrogen peroxide during the oxidation of benzylamine. The reaction was performed in 50 mM HEPES, pH 7.5 (assay buffer), initiated by addition 50 uL of various concentrations of AOC3 (diluted by assay buffer) to 50 ul substrate mixture of 2 mM Benzylamine, 2 units/mL HRP and 100 uM AUR. product-image-AAA161884_BIOACTIVITY13.jpg Bioactivity (Apolipoprotein M (APOM) is an approximately 25 kDa variably glycosylated protein that adopts a beta-barrel structure characteristic of lipocalin family proteins. It functions as a component of lipoprotein particles which play essential roles in fatty acid and cholesterol transport and metabolism. Alternative splicing generates a short isoform that lacks the N-terminal 72 amino acids. APOM is produced primarily by hepatocytes but also by renal tubule epithelial cells. The signal peptide is not cleaved and is required for APOM association with lipoprotein particles as well as Megalin mediated reabsorption by the kidney. The activity of recombinant mouse APOM was measured by its ability to bind all-trans retinoic acid. The binding of retinoic acid results in the quenching of Trp fluorescence in APOM. APOM was diluted to 50 ug/ml in 50 mM Tris, 10 mM CaCl2, 150 mM NaCl, pH 7.5 (assay buffer) and the retinoic acid was diluted to 400, 200, 100, 30, 10, 3, 1 and 0.5 uM in 95% ethanol. product-image-AAA161795_BIOACTIVITY13.jpg Bioactivity (Parathyroid hormone (PTH) is an 84-amino-acid peptide hormone that is secreted by the parathyroid gland. PTH plays an osteogenic role by acting directly on mesenchymal stem cells, cells with an osteoblastic lineage, osteocytes, and T cells. It also participates as an osteoclast by indirectly acting on osteoclast precursor cells and osteoclasts and directly acting on T cells. PTH is major endocrine regulators of vitamin D metabolism. product-image-AAA161706_BIOACTIVITY13.jpg Bioactivity (Urokinase Plasminogen Activator (uPA), also known as u-plasminogen activator or urokinase, is a highly-specific serine protease from the peptidase S1 family that cleaves plasminogen to form plasmin making it a key player in the plasminogen activator (PA) system. Expression of uPA is minimal in normal cells but is increased several fold in tumor cells by extracellular stimuli elevated in cancer and corresponds to poor outcomes in several types of cancer. Therefore, uPA has been identified as an excellent target for therapeutic development through inhibition of protease activity or though inhibition of uPA-dependent signaling while in complex with uPA receptor (uPAR). The activity assay of uPA was measured by its ability to cleave a peptide substrate, N-carbobenzyloxy-Gly-Gly-Arg-7-amido-4-methylcoumarin (Z-GGR-AMC). product-image-AAA161721_BIOACTIVITY13.jpg Bioactivity (Annexin V (ANXA5) is a multifunctional protein that is highly expressed on the apical surfaces of syncytiotrophoblasts, and plays an important role in haemostatic regulations, maintaining blood fluidity of the placenta. Lower ANXA5 levels have been observed in M2/ANXA5 haplotype carrying chorion. The association found between the maternal carriage of the M2/ANXA5 haplotype and an elevated risk of IUGR and/or PE supports the hypothesis that carrier status of this haplotype and the consequently reduced placental ANXA5 expression might be responsible, at least partially, for the onset of these gestational vascular complications. Annexin V is a calcium-dependent phospholipid binding protein that can be used to bind Phosphatidylserine (PS) during an early apoptosis event where the PS becomes exposed at the cell surface. Jurkat cells were treated with 10 uM camptothecin for 4h, 2*105 cells which were resuspended in binding buffer were stained with 5 ug recombinant human Annexin V-FITC and 10 ul Propidium iodide (PI) for 20min in dark room temperature. The flow cytometry was used to detect the early apoptotic and late apoptotic of camptothecin-treated Jurkat cells (Figure 2), the combination of Annexin V-FITC and propidium iodide allows for the distinction between early apoptotic cells (Annexin V-FITC positive and propidium iodide negative), late apoptotic and/or necrotic cells (Annexin V-FITC and propidium iodide positive), and viable cells (unstained). Thus, the recombinant human Annexin V-FITC can bind Phosphatidylserine (PS) at early apoptosis of Jurkat.)

The result was shown in figure 1, It was obvious that rhC5a can significantly induce N-acetyl-beta-D-glucosaminidase release.)

What Are Active Proteins?

Proteins are large molecules made up of long chains of amino acids.
They will typically fold into a very particular 3-dimensional shape/conformation, that is sometimes referred to as their “native” form, which allows them to work properly in the body. For the purposes of product categorization, AAA Biotech will typically refer to proteins purified from their original animal host as being “native” proteins (this is to signify their difference compared to their “recombinant” or “synthetic” protein counterparts).

If a protein successfully folds into the correct shape, it is will typically display high fidelity characteristics to its original protein in its original animal host, and be classified as an active protein, as it will be able to function “normally” in most enzymatic or binding capacities. If it loses this shape, due to factors such as heat or strong chemicals (such as detergents), it becomes inactive and is no longer able to perform its basic functions. All of the proteins in this category are made under strict quality control, and they are active, pure, low in contaminants, and stable.
Most are stored as freeze-dried powders and come without extra tags, so they’re very close to the actual natural/native form.

FAQ

1. What are active proteins used for in research?

Active proteins are used primarily in the study of how proteins function, in characterizing/discovering drug interactions, supporting cell growth, running biochemical assays, and in development of diagnostics or therapeutics.

2. How are AAA Biotech's active proteins validated?

AAA Biotech’s active proteins are validated through strict quality control and functional assays to ensure they are properly folded and active. “Active”, though, can be an ambiguous term, so if a specific “activity” or “binding” capability of a protein is of crucial interest to you, please inquire with us prior to purchase, and we will provide further details on how the “Active” modifier was determined to be applicable.

3. Are these proteins tested for biological activity?

Yes, all active proteins from AAA Biotech are tested to confirm they have the expected biological activity before being offered for use. Though, said “biological activity” can be either “enzymatic”, “binding”, or both.
