product-image-AAA150076_BIOACTIVITY10.jpg Bioactivity (Figure. The binding activity of TIMP3 with MMP2.Tissue Inhibitors Of Metalloproteinase 3 (TIMP3) is an protein belongs to the tissue inhibitor of metalloproteinases family. They are inhibitors of the matrix metalloproteinases. TIMP-3 is the only member of the TIMP family which is found exclusively in the extracellular matrix (ECM). It is regulated in a cell cycle-dependent fashion in certain cell types and may serve as a marker for terminal differentiation. Besides, Matrix Metalloproteinase 2 (MMP2) has been identified as an interactor of TIMP3, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse TIMP3 and recombinant mouse MMP2. Briefly, TIMP3 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100ul were then transferred to MMP2-coated microtiter wells and incubated for 2h at 37. Wells were washed with PBST and incubated for 1h with anti-TIMP3 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of TIMP3 and MMP2 was shown in Figure 1, and this effect was in a dose dependent manner.)

Myostatin is a secreted growth differentiation factor that produced and released by myocytes. This protein negatively regulates skeletal muscle cell proliferation and differentiation. Besides, Bone Morphogenetic Protein 1 (BMP1) has been identified as an interactor of MSTN, thus a binding ELISA assay was conducted to detect the interaction of recombinant human MSTN and recombinant human BMP1. Briefly, MSTN were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to BMP1-coated microtiter wells and incubated for 2h at 37. product-image-AAA150147_AD11.jpg Application Data (Semaphorin-3F (SEMA3F) is a member of semaphorins family. All the family members have a secretion signal, a 500-amino acid sema domain, and 16 conserved cysteine residues. Semaphorins not only guide axons in development, but also have major roles in immune function (classes 4, 6, and 7) and the development of bones. Class 3 semaphorins are one of the most versatile semaphorin classes, in which Sema3a is the most studied. Besides, SEMA3F is deleted or downregulated in many metastatic tumors. To test the effect of SEMA3F on inhibit cell proliferation, human lung carcinoma cells A549 were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach, replaced with serum-free overnight, then the medium was replaced with 2% serum standard DMEM prior to the addition of various concentrations of recombinant human SEMA3F. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37. Proliferation of A549 cells after incubation with SEMA3F for 48h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 assay after incubation with recombinant SEMA3F for 48h. The result was shown in Figure 2. It was obvious that SEMA3F significantly increased cell viability of A549 cells. (A) A549 cells cultured in DMEM, stimulated with 100ng/mL SEMA3F for 48h; (B) Unstimulated A549 cells cultured in DMEM for 48h.Figure. product-image-AAA152181_BIOACTIVITY13.png Bioactivity (Perforin 1 (PRF1) is a pore forming cytolytic protein found in the granules of cytotoxic T lymphocytes (CTLs) and NK cells. Upon degranulation, perforin binds to the target cell's plasma membrane, and oligomerises in a Ca2 dependent manner to form pores on the target cell. The pore formed allows for the passive diffusion of a family of pro-apoptotic proteases, known as the granzymes, into the target cell. The activity of recombinant PRF1 was measured by lysis of erythrocytes using a hemolysis assay. A general procedure is as fllows: two-fold dilute the recombinant mouse PRF1 with 0.9% NaCl, add 50ul a serial dilution of PRF1, 10ul 0.1M CaCl2 to each well, then add 50ul 0.25% rabbit erythrocyte (RaE) to each well and mixed gently. Add 50ul 0.9% NaCl to replace PRF1 in control wells. The plate is incubated for 20 hours at 37 degree C, 5% CO2. The results are shown in Figure 1. It was obvious that the minimal effective concentration of PRF1 is 5ug/ml.)

The protein is known to form disulfide-linked homodimers, but it can also be secreted as an oligomer. Fibulin 2 is considered a multifunctional binding protein due to its association with numerous ECM components, but its specific interactions have yet to be determined. Fibulin 2 is localized at the interface between microfibrils and the elastin core and its known interactions include nidogen-1, perlecan, laminin, aggrecan, endostatins, versican, collagen, and tropoelastin. It is down-regulated in numerous forms of cancer including breast, colorectal, lung, esophageal, and squamous cell carcinoma, but over-expression has been shown with solid tumors. To test the effect of FBLN2 on cell apoptosis, A549 cells were seeded into triplicate wells of 96-well plates at a density of 4,000 cells/well and allowed to attach overnight, then the medium was replaced with various concentrations of recombinant rat FBLN2 diluted with 5% serum standard DMEM. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10 ul of CCK-8 solution was added to each well of the plate, then the absorbance at 450 nm was measured using a microplate reader after incubating the plate for 1h at 37 degree C. Proliferation of A549 cells after incubation with FBLN2 for 72h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8) assay after incubation with recombinant rat FBLN2 for 72h. The result was shown in Figure 2. It was obvious that FBLN2 significantly inhibit cell viability of A549 cells. The ED50 is 0.97 ug/mL.)

product-image-AAA161865_BIOACTIVITY13.jpg Bioactivity (Complement Factor D (CFD) is a serine protease that catalyzes the initial proteolytic step in the alternative pathway of complement. Expressed in adipose tissue at high levels, factor D is also known as adipsin. It is an exceptionally specific protease and the only known protein substrate is factor B in complex with C3. Factor D protease activity is regulated by reversible conformational changes, which differs from the majority of serine proteases whose regulation involves either activation by processing of the zymogens or inactivation by binding of the inhibitors. Compared to its physiologically important proteolytic activity, factor D has much lower activity toward synthetic peptide substrates. However, thioester substrates have been routinely used for assessing factor D activity. The full-length (amino acid residues 1-263) of rat CFD was expressed which activity was measured by its ability to cleaves a thioester substrate Z-Lys-SBzl•HCl. The reaction was performed in 50 mM Tris, 1 M NaCl, pH 7.5 (Assay Buffer), initiated by addition 50 uL of various concentrations of CFD (diluted by Assay Buffer) to 50 ul substrate mixture of 0.2mM Z-Lys-SBzl•HCl and 0.2 mM DTNB. The final well serves as a negative control with no CFD, replaced with 50 ul assay buffer. Then read in kinetic mode for 5 minutes at an absorbance of 405 nm. The specific activity of recombinant rat CFD is > 10000 pmol/min/ug.)

What Are Active Proteins?

Proteins are large molecules made up of long chains of amino acids.
They will typically fold into a very particular 3-dimensional shape/conformation, that is sometimes referred to as their “native” form, which allows them to work properly in the body. For the purposes of product categorization, AAA Biotech will typically refer to proteins purified from their original animal host as being “native” proteins (this is to signify their difference compared to their “recombinant” or “synthetic” protein counterparts).

If a protein successfully folds into the correct shape, it is will typically display high fidelity characteristics to its original protein in its original animal host, and be classified as an active protein, as it will be able to function “normally” in most enzymatic or binding capacities. If it loses this shape, due to factors such as heat or strong chemicals (such as detergents), it becomes inactive and is no longer able to perform its basic functions. All of the proteins in this category are made under strict quality control, and they are active, pure, low in contaminants, and stable.
Most are stored as freeze-dried powders and come without extra tags, so they’re very close to the actual natural/native form.
